目的:建立HPLC法測定果汁中9種有機酸的檢測方法。

果汁中9種有機酸的反相高效液相色譜法測定
方法:果汁樣品經稀釋過濾后,采用Gemini C18色譜柱(4.6 mm×250 mm,5μm)分離,以0.01 mol/L磷酸氫二銨溶液(用磷酸調節pH=2.6)作為流動相,UV210 nm檢測,外標法定量。結果:9種有機酸的線性范圍分別為草酸1.0~400 mg/L、酒石酸2.0~500 mg/L、丙酮酸1.0~500 mg/L、蘋果酸5.0~2000 mg/L、抗壞血酸1.0~400 mg/L、乙酸8.0~2000 mg/L、檸檬酸5.0~2000 mg/L、富馬酸0.5~100 mg/L、反烏頭酸1.0~400 mg/L,各種有機酸的線性相關系數r>0.999,回收率在93.7%~109.0%,RSD均小于5.0%,最低檢測限在0.2~7.5 mg/L范圍。結論:該方法具有快速、靈敏、簡便、準確等優點,可應用于果汁中有機酸的檢測,能滿足實際工作的需要。
